Cholesterol is synthesised de novo within the liver by a series of reactions which utilize acetyl CoA substrate. The key enzyme in the pathway is HMG-CoA reductase. This is inhibited by cholesterol, so providing a means of regulating flux by negative feedback.
The pathway can be modified to advantage by HMG-CoA reductase inhibitors.
