Trimethoprim inhibits the enzyme dihydrofolate reductase which is involved in the conversion of folic acid to folinic (tetrahydrofolic) acid. The inhibition of the enzyme 50000 times greater in bacteria than it is in Man, so trimethoprim is relatively safe, though prolonged use with full doses can interfere with haemopoiesis.
