Drug interaction
Drug interactions should be thought of particularly carefully when the toxic/therapeutic ratio is low, as with:
- warfarin
- phenytoin
- oral hypoglycaemics
- oral contraceptives
- antihypertensives
- drugs acting on the brain
Mechanisms of drug action include:
- pharmaceutical
- pharmacokinetic: - absorption - distribution
- protein binding
- tissue binding - elimination - hepatic, renal - pharmacodynamic - direct and indirect
