As most of the enzymes in the pathway of gluconeogenesis are near equilibrium and unsaturated, the supply of substate has an important influence on the rate of glucose formation. For example, the rate may be proportional to lactate release from anaerobic glycolysis in muscle or glycerol release from adipocyte lipolysis.
