Drug interactions should be thought of particularly carefully when the toxic/therapeutic ratio is low, as with:

  • warfarin
  • phenytoin
  • oral hypoglycaemics
  • oral contraceptives
  • antihypertensives
  • drugs acting on the brain

Mechanisms of drug action include:

  • pharmaceutical
  • pharmacokinetic: - absorption - distribution
    • protein binding
    • tissue binding - elimination - hepatic, renal - pharmacodynamic - direct and indirect
